While properly validated and performant biomarkers are undoubtedly of immense value in medical decision-making, their discovery faces various hurdles. At a very practical level, for example, the tissue containing informative molecules needs to be accessible for sampling. Assays need to reach high levels of sensitivity, specificity, accuracy and reproducibility. Meeting these requirements is even more challenging in the context of diseases with high inter- and intra-patient variability or for samples with dynamic changes of cellular composition, such as blood or inflamed skin.
