Fragment-based drug discovery (FBDD) involves screening a small collection of very small compounds against a target molecule, and then optimizing and linking fragment “hits” to create high-affinity ligands for further development. FBDD forms the basis of many of today’s drug discovery programs because it requires smaller compound libraries, is likely to generate a higher number of hits than traditional high-throughput screening and fragments have been used successfully as a starting point for several approved drugs.
